本文报告普鲁卡因、加热、普鲁卡因联合加热对15例急性非淋巴细胞白血病患者骨髓的白血病祖细胞和正常骨髓粒单祖细胞的作用。普鲁卡因联合加热可以显著增强对白血病祖细胞的抑制作用,对正常人粒单祖细胞影响较小。单独应用42℃水浴加热1小时后作细胞培养,白血病祖细胞集落形成单位残存1/5左右;用2mM普鲁卡因则其残存不到1/20。后者明显提高了对白血病祖细胞的体外杀伤效果。而后,我们对1例急性早幼粒细胞白血病患者的骨髓用后一方法体外净化并进行移植,患者骨髓回输无任何不适反应,造血功能恢复良好。
This article reports the combined effects of procaine, heating and procaine on leukemic progenitor cells and normal myeloid progenitor cells from 15 patients with acute non-lymphocytic leukemia. Procaine combined with heating can significantly enhance the inhibition of leukemia progenitor cells, normal human granulocyte progenitor cells less affected. The cells were cultured in a 42 ° C water bath alone for 1 hour, and the leukemic progenitor colony forming units remained at about 1/5. However, 2 mM procaine remained less than 1/20. The latter significantly enhances the in vitro killing of leukemic progenitor cells. Then, we in one case of acute promyelocytic leukemia in patients with a method of bone marrow purification and transplantation in vitro, patients without any discomfort of bone marrow transfusion, hematopoietic function recovered well.
