目的:研究脑舒胶囊对VD模型大鼠的中枢免疫炎症反应的抑制作用。方法:反复夹闭双侧颈总动脉加硝普钠降压复制VD大鼠模型;Morris水迷宫检测大鼠学习记忆力;放射性免疫法测定血清IL-6含量;ELISA法测定血清sIL-2R含量;免疫组织化学检测TNF-α阳性表达。结果:与假手术组比较,模型组大鼠血清IL-6及sIL-2R含量明显增加,与模型组相比,0.36g/kg、0.72g/kg、1.44g/kg脑舒胶囊均可降低大鼠血清IL-6及sIL-2R含量,具有一定剂量依赖性。光镜下(×400)下,模型组大鼠海马CA1区胞浆及胞膜棕褐色颗粒明显增加,各剂量脑舒胶囊治疗组大鼠海马CA1区胞浆及胞膜棕褐色颗粒明显减少;与假手术组比较,模型组TNF-α阳性反应的平均光密度(ID)均明显降低,各治疗组TNF-α阳性反应的ID均下降,且有一定剂量依赖性。结论:脑舒胶囊对VD大鼠中枢免疫炎症反应有较好抑制作用。
Objective: To study the inhibitory effect of Naoshu capsule on central immune inflammation in VD model rats. Methods: The bilateral common carotid arteries were implanted with antihypertensive sodium nitroprusside to replicate the VD model. Morris water maze was used to measure the learning and memory ability of rats. The level of IL-6 in serum was measured by radioimmunoassay. The level of sIL-2R in serum was measured by ELISA. Immunohistochemistry was used to detect the positive expression of TNF-α. Results: Compared with the sham operation group, the contents of IL-6 and sIL-2R in the model group were significantly increased, compared with the model group, 0.36g / kg, 0.72g / kg and 1.44g / kg Naoshu capsule Serum levels of IL-6 and sIL-2R in rats were dose-dependent. Under the light microscope (× 400), the cytoplasm and membrane tan particles in the hippocampal CA1 region of the model group were significantly increased, and the cytoplasm and the membrane tan granules of the hippocampal CA1 region were significantly decreased in each dose of Naoshu capsule. Compared with the sham-operation group, the average optical density (ID) of TNF-αpositive reaction in the model group was significantly decreased, and the ID of TNF-αpositive reaction in each treatment group was decreased in a dose-dependent manner. Conclusion: Naoshu Capsule has a good inhibitory effect on central immune-inflammatory reaction in VD rats.
