腺苷酸类是参与遗传信息贮存、传递与表达的核酸“构件”分子,也是生物氧化、能量代谢中的能源物质,ATP 又是临床治疗脑血管意外、冠心病及心肌梗塞等的药物。因此对生物组织或体液中的游离腺苷酸类寻求简便、快速、灵敏的测定方法具有重要意义。我们采用自装“Permaphase AAX”的不锈钢柱2.1mm×100cm,理论塔板数为1110/米,流动相为0.005M 磷酸二氢钾(pH3.0)和0.5M 磷酸二氢钾(pH4.0),每分钟以3%线性浓度梯度洗脱。样品柱和参比柱的洗脱液流速分别是0.64ml/分和0.76ml/分,柱压50kg/cm~2,柱温为室温,记录器纸速10mm/分,检测敏度0.08AVFS,检测波长257nm。在上述条件下,三种标准腺苷酸混合溶液保留时间分别为AMP tr=2分44秒、ADP
