目的研究华蟾素注射剂在小鼠体内的最大耐受量以及不同剂量华蟾素对荷人胰腺癌细胞株SW1990裸小鼠的抑瘤作用。方法将华蟾素分为100、75、50、30、10、3 g·kg~(-1)共6个不同剂量组并设对照组(生理盐水组),观察该药对CD1小鼠的最大耐受量;根据最大耐受量的结果将荷人胰腺癌细胞株SW1990皮下移植瘤裸小鼠,之后随机分为对照组与吉西他滨组,以及华蟾素12.5、25、50 g·kg~(-1)剂量组,定期测量肿瘤体积,给药21 d后计算抑瘤率。结果华蟾素毒性反应出现在100、75 g·kg~(-1)两剂量组。华蟾素小剂量组抑瘤率为16.95%,中剂量组为30.51%,大剂量组为18.64%,各给药组的平均瘤体积均小于对照组(P<0.05);其中华蟾素中剂量组的抑瘤率较好。华蟾素各剂量组裸小鼠均生长良好,无明显毒性反应。结论华蟾素在CD1小鼠体内的最大耐受剂量为50 g·kg~(-1),华蟾素给药剂量为25 g·kg~(-1)时对荷人胰腺癌细胞株SW1990裸小鼠抑瘤作用最佳。
Objective To study the maximum tolerability of cinobufagin injection in mice and the antitumor effects of cinobufacini on nude mice bearing human pancreatic cancer cell line SW1990. Methods Cinobufacini was divided into 6 groups of 100, 75, 50, 30, 10, 3 and 3 g · kg ~ (-1), and the control group (NS group) According to the maximum tolerated dose, the human pancreatic cancer cell line SW1990 was subcutaneously transplanted into nude mice and then randomly divided into control group and gemcitabine group, and cinobufacine 12.5, 25 and 50 g · kg ~ (-1) dose group. The tumor volume was measured regularly, and the inhibition rate was calculated after 21 days. Results The cinobufotalin toxicity appeared in two doses of 100 and 75 g · kg -1. The inhibitory rate of low dose cinobufotalin group was 16.95%, middle dose group was 30.51%, and high dose group was 18.64%. The mean tumor volume of each group was less than that of the control group (P <0.05) The dose group of tumor inhibition rate is better. Cinobufagin all doses of nude mice grew well, no significant toxicity. Conclusions The maximum tolerated dose of cinobufacini in CD1 mice is 50 g · kg -1, and when the concentration of cinobufagin is 25 g · kg -1, the effect on the growth of human pancreatic cancer cell line SW1990 Tumor inhibition of nude mice best.
