目的初步探讨匝迪-5味丸对抑郁模型大鼠行为学及学习记忆能力的影响,评价其抗抑郁作用及对学习记忆的干预作用。方法 SD雄性大鼠,根据体质量随机分为正常组、模型组、氟西汀组(3.6 mg/kg),匝迪-5味丸低、中、高剂量组(0.2、0.4、0.8 mg/kg),每组10只。采用应激结合孤养的方法复制抑郁模型,以矿场(Open-field)实验、糖水消耗实验及体质量变化来进行行为学评价,应用Morris水迷宫试验来检测大鼠学习记忆能力。结果给予21d应激后模型大鼠糖水消耗量减少,体质量增长缓慢,Open-Field实验水平活动得分和垂直活动得分及Morris水迷宫实验逃避潜伏期较正常组显著降低(P<0.05,P<0.01)。与模型组相比,氟西汀和匝迪-5味丸高、中剂量组可不同程度提高模型大鼠糖水消耗量、Open-Field实验水平活动和垂直活动及学习记忆能力(P<0.05,P<0.01)。结论匝迪-5味丸具有一定的抗抑郁作用,其机制值得进一步研究。
Objective To investigate the effect of Zodioxin-5 taste pill on the behavior and learning and memory abilities of rats with depression and to evaluate its antidepressant effect and the intervention effect on learning and memory. Methods SD male rats were randomly divided into normal group, model group, fluoxetine group (3.6 mg / kg), low, medium and high doses of ZGD group (0.2, 0.4 and 0.8 mg / kg), 10 in each group. The stress model was used to replicate the depression model. The behavioral evaluation was carried out by using Open-field experiment, sugar consumption experiment and body weight change. Morris water maze test was used to detect the learning and memory ability of rats. Results After 21 days of stress, the consumption of syrup and the body weight of model rats decreased slowly. The score of Open-Field test and vertical activity score and the escape latency of Morris water maze test were significantly lower than those of normal group (P <0.05, P <0.01) ). Compared with the model group, fluoxetine and Zizhi-5 Weiwan high and medium dose groups could increase the sugar consumption, Open-Field experimental activity and vertical activity and learning and memory ability of the model rats to varying degrees (P <0.05, P <0.01). Conclusion Resi 5 taste pill has certain antidepressant effect, its mechanism deserves further study.
