目的:研究通痹合剂对佐剂型关节炎大鼠软骨细胞产生保护作用的机制。方法:培养正常大鼠软骨细胞,大鼠白介素(IL-1β)刺激后用ELISA法及阿利新蓝法检测10%含药血清对各组大鼠软骨细胞产生蛋白多糖(GAG)及Ⅱ型胶原的含量的影响。结果:通痹合剂组、消炎痛组、模型组佐剂型关节炎模型(AA)大鼠软骨细胞产生Ⅱ型胶原与正常组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组与正常组比较,差异无显著性意义(P>0.05)。通痹合剂组、消炎痛组产生Ⅱ型胶原与模型组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组、模型组及通痹合剂组与柳氮磺胺吡啶组分别比较,差异均无显著性意义(P>0.05);消炎痛组与柳氮磺胺吡啶组比较,差异有显著性意义(P<0.05)。通痹合剂组、消炎痛组、模型组AA大鼠软骨细胞产生GAG与正常组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组与正常组比较,差异无显著性意义(P>0.05)。通痹合剂组、消炎痛组产生GAG与模型组比较,差异均有非常显著性意义(P<0.01);柳氮磺胺吡啶组、模型组、通痹合剂组与柳氮磺胺吡啶组分别比较,差异均无显著性意义(P>0.05);消炎痛组与柳氮磺胺吡啶组比较,差异有显著性意义(P<0.05)。结论:通痹合剂对软骨细胞有保护作用,机制可能与其增加Ⅱ型胶原与GAG含量有关。
Objective: To study the mechanism of Tongbai Mixture on chondrocytes in adjuvant arthritis rats. Methods: The normal rat chondrocytes were cultured. Tongbi Mixture group, indomethacin group produced GAG compared with the model group, the difference was significant (P <0.01); sulfasalazine group, model group, Tongbi Mixture group and sulfasalazine group were compared, There was no significant difference (P> 0.05) between indomethacin group and sulfasalazine group (P <0.05). Conclusion: Tongbi Mixture has a protective effect on chondrocytes, which may be related to the increase of type Ⅱ collagen and GAG content.
