目的:研究丹参酮ⅡA(TanⅡA)联合顺铂(DDP)对咽鳞癌Fadu细胞的肿瘤抑制作用及其机制。方法:利用CCK-8法检测TanⅡA联合DDP对Fadu细胞增殖抑制率的影响,流式细胞分析法检测细胞凋亡率变化及细胞周期分布情况,Western blotting法检测survivin、cleaved caspase-3及cleaved PARP蛋白的表达水平。结果:与丹参酮ⅡA和顺铂单独作用相比,两药联合作用于下咽鳞癌Fadu细胞72h后,细胞增殖受到明显抑制(均P<0.01),细胞周期阻滞于S期,凋亡细胞比例增加显著;caspase-3和PARP剪切段蛋白表达增多,survivin蛋白表达明显减少(P<0.01)。结论:丹参酮ⅡA联合顺铂具有协同抗咽鳞癌Fadu细胞的作用,其作用机制可能与下调凋亡抑制蛋白survivin的表达有关。
Objective: To study the tumor inhibitory effect of Tan Ⅱ A combined with cisplatin on pharynx squamous cell carcinoma Fadu cells and its mechanism. Methods: The effect of TanⅡA combined with DDP on the proliferation inhibition rate of Fadu cells was detected by CCK-8 assay. The apoptosis rate and cell cycle distribution were detected by flow cytometry. The expressions of survivin, cleaved caspase-3 and cleaved PARP were detected by Western blotting Protein expression levels. Results: Compared with tanshinone ⅡA and cisplatin, the proliferation of Fadu cells was significantly inhibited (all P <0.01) after treated with both drugs for 72 hours, the cell cycle arrest was in S phase and apoptotic cells (P <0.01). The expressions of caspase-3 and PARP in the cleaved segment increased and the expression of survivin protein decreased significantly (P <0.01). CONCLUSION: Tanshinone IIA combined with cisplatin has the synergistic effect on Fadu cells induced by anti-pharyngeal squamous cell carcinoma. Its mechanism may be related to the down-regulation of survivin protein.
