目的 探讨热休克蛋白 (HSP 70 )反义寡核苷酸逆转人前列腺癌细胞株LNCaP及PC 3m耐药性的作用。 方法 用MTT法和成集落试验测定了 10 μmol/LHSP 70反义寡核苷酸作用后不同浓度阿霉素、丝裂霉素C对LNCaP和PC 3m生长抑制率的影响。 结果 10 μmol/LHSP 70反义寡核苷酸可以显著增加 4~ 12 μg/ml阿霉素和 10~ 10 0 μg/ml丝裂霉素 C对PC 3m的生长抑制率以及 0 .0 1~ 0 .0 8μg/ml阿霉素和 1~ 6 μg/ml丝裂霉素C对LNCaP的生长抑制率 ,差别有显著性意义 (P <0 .0 5 )。抑制率均超过 5 0 %。 结论 HSP 70与前列腺癌细胞的耐药性有关 ;HSP 70反义寡核苷酸可以特异性地逆转二种不同生物学特性的前列腺癌细胞的耐药性 ,为临床前列腺癌化疗提供了新方法
Objective To investigate the effects of heat shock protein (HSP 70 ) antisense oligonucleotides on the reversal of drug resistance of human prostate cancer cell lines LNCaP and PC 3m. Methods The effects of different concentrations of doxorubicin and mitomycin C on the growth inhibition of LNCaP and PC 3m after 10 μmol/L HSP 70 antisense oligodeoxynucleotides were determined by MTT assay and colony forming assay. Results 10 μmol/L HSP 70 antisense oligodeoxynucleotide significantly increased the inhibition rate of PC 3m and the inhibition of PC 3m by 4~12 μg/ml adriamycin and 10-100 μg/ml mitomycin C. The growth inhibition rate of LNCaP between 0. 0 8μg/ml adriamycin and 1~6 μg/ml mitomycin C showed significant difference (P < 0.05). The inhibition rate exceeded 50%. Conclusion HSP 70 is related to the drug resistance of prostate cancer cells; HSP 70 antisense oligonucleotides can specifically reverse the resistance of two different biological characteristics of prostate cancer cells, providing a new method for clinical prostate cancer chemotherapy.
