1型糖尿病主要是由于胰岛β细胞破坏导致胰岛素绝对缺乏引起,胰岛β细胞无法再生,也不能进行异种移植,干细胞移植是治疗糖尿病的新型方法。已有研究证实了脂肪间充质干细胞(adipose mesenchymal stem cells,ADSC)能够被诱导分化成胰岛素分泌细胞,也有研究显示了胰十二指肠同源盒基因-1(PDX-1)可诱导非胰岛分泌细胞如骨髓间充质干细胞、脂肪间充质干细胞等向胰岛素分泌细胞分化,提示PDX-1在改善胰岛β细胞再生和促进胰岛细胞恢复中有着巨大潜能。本文就当前PDX-1基因提高脂肪间充质干细胞分化为胰岛分泌细胞能力的研究前景作一综述。
Type 1 diabetes is caused mainly by the absolute lack of insulin due to islet beta cell destruction, islet beta cells can not be regenerated, and xenografts can not be performed. Stem cell transplantation is a novel method for the treatment of diabetes. It has been confirmed that adipose mesenchymal stem cells (ADSCs) can be induced to differentiate into insulin-secreting cells. Other studies have shown that pancreatic duodenal homeobox gene-1 (PDX-1) Islet secreting cells such as bone marrow mesenchymal stem cells and adipose-derived mesenchymal stem cells differentiate into insulin-secreting cells, suggesting that PDX-1 has great potential in improving islet β-cell regeneration and promoting islet cell recovery. This review summarizes the research prospects of the current PDX-1 gene in enhancing the ability of adipose-derived mesenchymal stem cells to differentiate into islet-secreting cells.
