目的探讨蛋白酪氨酸磷酸酶SHP-1 mRNA在γ射线体外诱发的白血病小鼠肿瘤细胞中的变化情况,为进一步从细胞信号转导途径入手研究辐射致癌的分子机制奠定一定的基础。方法实验分为癌变组、未癌变组及对照组,应用荧光定量PCR法分别检测各组胸腺及骨髓细胞中SHP-1 mRNA的含量变化情况。结果癌变组胸腺细胞中SHP-1 mRNA的含量与未癌变组及对照组比无明显差异,而骨髓细胞中SHP-1 mRNA的含量明显高于对照组。结论在γ射线诱发的白血病小鼠,胸腺细胞SHP-1 mRNA表达无明显变化,但存在SHP-1的翻译异常,表现为翻译增强现象,骨髓细胞中SHP-1 mRNA的表达明显增强。
Objective To investigate the changes of SHP-1 mRNA in tumor cells of leukemia mice induced by γ-rays in vitro, and to lay a foundation for the further study of the molecular mechanism of radiation-induced cancer by signal transduction pathway of cells. Methods The experiment was divided into cancerous group, non-cancerous group and control group. Fluorescence quantitative PCR was used to detect the content of SHP-1 mRNA in thymus and bone marrow cells. Results There was no significant difference in SHP-1 mRNA expression in thymocytes of cancerous group compared with that in non-cancerous group and control group, while the content of SHP-1 mRNA in bone marrow cells was significantly higher than that in control group. Conclusions There is no obvious change of SHP-1 mRNA expression in thymocytes in γ-ray-induced leukemia mice, but there is an abnormal translation of SHP-1. The SHP-1 mRNA expression is obviously enhanced in bone marrow cells.
