以药用植物金银花为材料,对其根部、茎部、叶片不同部位的内生细菌进行分离纯化,对细胞特征采用革兰氏染色、芽孢染色法进行研究,结合菌落特征和生理生化特性将其内生细菌初步分为第1类型和第2类型两大类。采用稀释涂布法对金银花不同部位分别进行可培养内生细菌计数。结果表明,内生菌的总体数量分布为根部>叶片>茎部,说明内生细菌在金银花组织中的分布因部位不同而不同。基于细胞染色特征、显微观察结果和培养特性,内生菌中第1类型符合芽孢杆菌属(Bacillus)的特点,第2类型具有类芽孢杆菌属(Paenibacillus)的特征。
The medicinal plant honeysuckle was used as material to isolate and purify the endophytic bacteria in different parts of roots, stems and leaves. Gram stain and spore staining were used to study the cell characteristics. Combining the characteristics of colony and physiological and biochemical characteristics, Endophytic bacteria initially divided into two types of type 1 and type 2. Using dilution coating method honeysuckle different parts were cultured endophytic bacteria count. The results showed that the total number of endophyte was root> leaf> stem, indicating that the distribution of endophytic bacteria in honeysuckle tissue varies with different parts. Based on the characteristics of cell staining, microscopic observation and culture characteristics, the first type of endophytic bacteria is characterized by Bacillus and the second type is characterized by Paenibacillus.
