目的:探讨普鲁卡因(procaine)对人鼻咽癌(NPC)细胞CNE-2Z的影响。方法:体外培养的NPC细胞CNE-2Z分别加入不同浓度的普鲁卡因,并作用不同的时间。应用四甲基偶氮唑盐[3-(4,5-Dimethyl-thiaoly)-2,5-diphenyl-2H tetrazolium bromide,MTT]法和流式细胞仪检测观察普鲁卡因对NPC细胞CNE-2Z的影响,同时用RT-PCR分析p16和RASSF1A mRNA的表达情况。结果:普鲁卡因能显著抑制CNE-2Z细胞增殖,该作用呈剂量-效应和时间依赖关系。普鲁卡因使细胞周期阻滞于G1期,并能上调CNE-2Z细胞RASSF1A mRNA的表达,但对p16 mRNA表达无明显影响。结论:普鲁卡因对CNE-2Z细胞有增殖抑制作用,它能上调CNE-2Z细胞RASSF1A mRNA的表达,推测这可能是普鲁卡因抑制CNE-2Z细胞增殖的重要分子机制之一。
Objective: To investigate the effect of procaine on CNE-2Z in human nasopharyngeal carcinoma (NPC) cells. Methods: CNE-2Z cells cultured in vitro were added with different concentrations of procaine, and the role of different time. The effect of procaine on the expression of CNE-2 in NPC cells was analyzed by MTT assay and flow cytometry (FCM) 2Z at the same time using RT-PCR analysis p16 and RASSF1A mRNA expression. Results: Procaine significantly inhibited the proliferation of CNE-2Z cells in a time-and dose-dependent manner. Procaine blocked the cell cycle in G1 phase and up-regulated the expression of RASSF1A mRNA in CNE-2Z cells, but had no effect on the expression of p16 mRNA. CONCLUSION: Procaine can inhibit the proliferation of CNE-2Z cells and up-regulate the expression of RASSF1A mRNA in CNE-2Z cells. This may be one of the important molecular mechanisms that procaine inhibits the proliferation of CNE-2Z cells.
