采用薄膜-超声分散法制备蛇床子素脂质体,以包封率为评价指标,采用单因素考察和正交试验优化蛇床子素脂质体的处方和工艺条件。所得优化工艺为:胆固醇与蛋黄卵磷脂质量比为1∶2,药脂比为3∶20,Tris缓冲液p H值为8.2,成膜温度为40℃。优化后3批脂质体的平均粒径和?电位为260 nm和-28.1 m V,包封率为88%。与蛇床子素溶液相比,蛇床子素脂质体的体外释放呈现出一定的缓释特征。
The osthole liposomes were prepared by the membrane-ultrasonic dispersion method. The entrapment efficiency was used as the evaluation index to optimize the formulation and processing conditions of the osthole liposomes by single factor test and orthogonal experiment. The optimized process was as follows: the mass ratio of cholesterol to egg yolk lecithin was 1: 2, the drug-lipid ratio was 3:20, the pH value of Tris buffer solution was 8.2, and the film-forming temperature was 40 ℃. After optimization, the average diameters and potentials of three batches of liposomes were 260 nm and -28.1 mV, respectively. The encapsulation efficiency was 88%. Compared with osthole solution, the release of osthole liposome in vitro showed some sustained release characteristics.
