目的 研究雌激素受体 (ER)基因多态性在 2型糖尿病 (DM )患者中的分布及其与血脂的关系。方法 应用聚合酶链反应(PCR)和限制性片段长度多态性 (RFLP)方法检测 13 1例 2型DM患者ERXbaI、PvuII酶切多态性基因型 ,分析其分布特点及其与血脂水平的关系。结果 2型DM患者中 ,以xx型 (62 .6% )、Pp型 (5 3 .4% )和xxpp型 (3 1.3 % )频率最高。男女间各基因型分布的差异无显著性意义 (P >0 .0 5 )。Xx型的血清TC、LDL -C、ApoB水平较xx型明显增高 (P <0 .0 1或 <0 .0 5 ) ;Pp型LDL -C水平较pp型显著增高 (P <0 .0 5 )。多元回归分析显示 ,TC水平分别与XbaI和PvuII多态突变型独立相关 (OR =1.64 5P =0 .0 0 7;OR =1.5 3 7P =0 .0 3 5 )。结论 2型DM患者ER基因突变可能导致血脂水平变化 ,该变化可能与大血管疾病 (MA)的发生有关。
Objective To investigate the distribution of estrogen receptor (ER) gene polymorphism in type 2 diabetes mellitus (DM) and its relationship with serum lipids. Methods The genotypes of ERXbaI and PvuII restriction digests were detected by polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) in 131 cases of type 2 diabetes mellitus (DM). The distribution characteristics of ERXbaI and PvuII were analyzed. relationship. Results Type 2 DM patients had the highest frequency of type xx (62.6%), type Pp (53.4%) and type xxpp (3 1.3%). There was no significant difference in genotype distribution between men and women (P> 0.05). The level of serum TC, LDL-C and ApoB in Xx group was significantly higher than that of xx group (P <0.01 or <0.05), while the level of Pp-type LDL-C was significantly higher than that in PP group ). Multivariate regression analysis showed that TC levels were independently associated with XbaI and PvuII polymorphisms (OR = 1.64 5P = 0.07; OR = 1.53 7P = 0.0305, respectively). Conclusion The mutation of ER gene in type 2 DM patients may lead to the change of blood lipid level, which may be related to the occurrence of macrovascular disease (MA).
