用生物素标记低密度脂蛋白(LDL),并建立了测定单个核细胞LDL受体活性的生物素-亲和素-酶联吸附测定法。应用该法测定了20例高胆固醇血症患者和55例正常人新鲜分离的单个核细胞LDL受体活性。结果显示:高胆固醇血症患者(血胆固醇浓度7.97±1.61mmol/L)的受体活性为50.1±16.0ng/mg细胞蛋白,正常人(血胆固醇浓度4.16±0.65mmol/L)的受体活性为79.1±34.6ng/mg细胞蛋白,两者比较P<0.01。这表明患者血胆固醇浓度的升高与其受体活性的降低有着密切的关系。BA-ELSA法用血量仅5ml/人,且敏感、简便、安全,适用于大规模临床检测。
Low density lipoprotein (LDL) was labeled with biotin, and biotin-avidin-ELISA assay was established to determine LDL receptor activity of mononuclear cells. The LDL receptor activity of freshly isolated mononuclear cells from 20 patients with hypercholesterolemia and 55 normal subjects was determined by this method. The results showed that the receptor activity of hypercholesterolemia patients (serum cholesterol concentration 7.97 ± 1.61mmol / L) was 50.1 ± 16.0ng / mg cell protein, and that of normal subjects (serum cholesterol concentration 4.16 ± 0.65mmol / L) 79.1 ± 34.6ng / mg cell protein, P <0.01. This shows that patients with elevated blood cholesterol levels and receptor activity is closely related to the decline. BA-ELSA method with only 5ml blood / person, and sensitive, simple and safe, suitable for large-scale clinical testing.
