目的探讨开窍中药冰片、牛黄、麝香、石菖蒲及苏合香对牛肾上腺髓质细胞儿茶酚胺分泌的影响。方法体外培养牛肾上腺髓质细胞,HPLC-ECD检测其分泌儿茶酚胺的含量,分别考察5种开窍药对乙酰胆碱、藜芦定碱和56 mmol·L-1高钾溶液诱导的儿茶酚胺分泌的影响。结果5种开窍中药均显著抑制乙酰胆碱、藜芦定碱诱导的儿茶酚胺分泌(P<0.05或P<0.01)而对高钾溶液诱导的儿茶酚胺分泌均无显著影响;对乙酰胆碱诱导的儿茶酚胺分泌的抑制作用随浓度升高而增强(P<0.05或P<0.01)。结论开窍中药具有抑制交感活性作用,且通过乙酰胆碱受体途径和电压依赖型钠离子通道发挥作用。
Objective To investigate the effects of Chinese herbal medicine (BLM), bezoar, rugosa, Shichangpu and Suhexiang on the secretion of catecholamines from bovine adrenal medulla cells. Methods Bovine adrenal medulla cells were cultured in vitro. The content of catecholamine excreted was determined by HPLC-ECD. The effects of five kinds of herbs on the secretion of catecholamines induced by acetylcholine, veratridine and 56 mmol·L-1 high potassium solution were investigated. RESULTS: All of the 5 kinds of Chinese herbal medicines significantly inhibited the catecholamine-induced catecholamine secretion induced by acetylcholine and veratridine (P<0.05 or P<0.01) but had no significant effect on the secretion of catecholamine induced by hyperkalemia solution; the inhibitory effect on acetylcholine-induced catecholamine secretion was observed. Increased with increasing concentrations (P<0.05 or P<0.01). Conclusion Herbal medicine with traditional Chinese medicine has the effect of inhibiting sympathetic activity, and plays a role through the acetylcholine receptor pathway and voltage-dependent sodium channels.
