目的研究不同白细胞与肿瘤细胞浓度比对剪切流环境中肿瘤细胞在内皮细胞上黏附过程的影响。方法在平行平板流动腔中加入不同浓度比的白细胞和肿瘤细胞,分析不同剪切率下(50、100、200 s-1)肿瘤细胞在内皮细胞上瞬时黏附和累积黏附的变化。结果细胞浓度比为3∶1时,肿瘤细胞在内皮细胞上的瞬时黏附和累积黏附均比细胞浓度比为1∶1时有显著提升,且在高剪切率(100、200 s-1)下更为明显。在观察的5 min内,随着剪切率的增加,细胞浓度比增大对肿瘤细胞黏附产生影响的时间点有所提前。结论在剪切流中白细胞浓度增加对肿瘤细胞在内皮细胞上的黏附起到促进作用,结果为研究肿瘤在血管中转移和靶向治疗提供有意义的探索。
Objective To study the effect of concentration ratios of leukocytes to tumor cells on the adhesion of tumor cells to endothelial cells in shear flow environment. Methods The leukocytes and tumor cells with different concentration ratios were added into the parallel plate flow chamber to analyze the changes of transient adhesion and accumulation of endothelial cells in different shear rates (50, 100, 200 s-1). Results When the cell concentration ratio was 3:1, the transient adhesion and accumulation of tumor cells on endothelial cells were significantly enhanced compared with the cell concentration ratio of 1: 1, and at high shear rate (100, 200 s-1) Under the more obvious. Within 5 min of observation, with the increase of the shear rate, the time point of the influence of the cell concentration ratio on tumor cell adhesion increased. Conclusions The increase of leukocyte concentration in shear flow can promote the adhesion of tumor cells to endothelial cells. The results provide a meaningful exploration of tumor metastasis and targeted therapy in blood vessels.
