目的探讨TGIF对TGF-β诱导胃癌细胞凋亡的影响。方法将pcDNA3.1-TGIF质粒稳定转染胃癌细胞BGC823细胞,然后以流式细胞术分析BGC823细胞凋亡率的改变,以western blot分析TGIF,caspase8和caspase9蛋白含量变化。结果TGF-β1作用后,BGC823细胞凋亡率明显增加,同时伴有caspase9的激活,但不伴caspase8的激活;与对照组相比,pcDNA3.1-TGIF质粒转染细胞对TGF-β诱导的细胞凋亡敏感性明显降低。结论TGF-β通过caspase9途径诱导BGC823细胞的凋亡;TGIF可抑制TGF-β诱导的细胞凋亡。
Objective To investigate the effect of TGIF on TGF-β-induced gastric cancer cell apoptosis. Methods The pcDNA3.1-TGIF plasmid was stably transfected into BGC823 cells. Then the apoptosis rate of BGC823 cells was analyzed by flow cytometry. The protein contents of TGIF, caspase8 and caspase9 were analyzed by western blot. Results After treatment with TGF-β1, the apoptosis rate of BGC823 cells was significantly increased, accompanied by the activation of caspase9, but not with the activation of caspase8. Compared with the control group, the transfection of pcDNA3.1-TGIF plasmid to TGF- Apoptosis sensitivity was significantly reduced. Conclusion TGF-β induces apoptosis in BGC823 cells through caspase9 pathway. TGIF inhibits TGF-β-induced apoptosis.
