从湖北省大冶市古铜矿区域分离得到1株具有较高Fe~(2+)氧化活性的嗜酸氧化亚铁硫杆菌SY,16S rDNA序列分析表明,该菌与Acidithiobacillus ferrooxidans(DQ 062116.1)的序列相似性最高。生理生化性质测定表明,菌株SY化能自养型,最适pH值为2.0,最适生长温度为30℃。菌株SY对Cu~(2+)、Cd~(2+)、Ni~(2+)、Zn~(2+)等重金属离子抗性的最低抑菌浓度分别为300、350、700和800 mmol/L,表明其对多种重金属离子均有很高的抗性。SY浸矿数据表明该菌对湖北大冶本土矿石浸出率为84.28%,高于其他地区矿石浸出率,在本土矿石生物浸出中显示出很大的优势。
A strain of Acidithiobacillus ferrooxidans SY, 16S rDNA isolated from the ancient copper mine in Daye City, Hubei Province, was sequenced. The results showed that the strain was highly sensitive to Acidithiobacillus ferrooxidans (DQ 062116.1) Sequence similarity is the highest. Physiological and biochemical properties showed that strain SY could autotrophic, the optimum pH value was 2.0 and the optimum growth temperature was 30 ℃. The minimum inhibitory concentrations of SY against heavy metal ions such as Cu 2+, Cd 2+, Ni 2+, Zn 2+ were 300, 350, 700 and 800 mmol, respectively / L, indicating that it has a high resistance to a variety of heavy metal ions. The leaching data of SY shows that the leaching rate of this bacterium to native ore in Daye Hubei is 84.28%, which is higher than the leaching rate of ore in other areas and shows great advantage in bioleaching of native ore.
