目的探索适合海藻糖和葡萄糖联合负载红细胞的时间,为冷冻干燥红细胞提供基础。方法分别采用0、0.125、0.25、0.5和1 mol/L的海藻糖联合葡萄糖,在37℃下负载红细胞4、6和8 h。然后检测负载红细胞内海藻糖和葡萄糖的浓度。结果随着负载时间的延长,进入红细胞内的海藻糖和葡萄糖数量增加。并且随着负载液浓度的增加,红细胞内的海藻糖和葡萄糖数量增加。负载时间为8 h时,进入红细胞的葡萄糖比6 h组明显增加。但是,延长负载时间为8 h后,进入红细胞的海藻糖浓度与6 h组比较,差异无统计学意义。结论海藻糖联合葡萄糖负载红细胞6 h后,进入红细胞内的海藻糖和葡萄糖,能够满足冰冻干燥红细胞的负载要求。
Objective To explore the suitable time for the combination of trehalose and glucose loaded erythrocytes to provide the basis for lyophilization of erythrocytes. Methods Treated with 0, 0.125, 0.25, 0.5 and 1 mol / L trehalose with glucose respectively, the erythrocytes were loaded at 37 ℃ for 4, 6 and 8 h. Then detect the concentration of trehalose and glucose in the loaded erythrocytes. Results As load time increased, the amount of trehalose and glucose entering the erythrocytes increased. And as the concentration of the loading fluid increases, the amount of trehalose and glucose in the erythrocytes increases. When the load time was 8 h, the glucose entering the erythrocytes significantly increased than the 6 h group. However, after prolonged loading for 8 h, the concentration of trehalose entering the erythrocytes was not significantly different from that of the 6 h group. Conclusion Trehalose and glucose loaded red blood cells 6 h, into the erythrocytes trehalose and glucose, to meet the load requirements of lyophilized red blood cells.
