目的考察重组降血压肽(recombinant antihypertensive peptide,rAHP,序列为VLPVPR)可生物降解聚(乳酸-羟基乙酸)共聚物(PLGA)缓释微球对自发性高血压大鼠(SHR)的降压作用。方法采用口服和皮下注射两种给药方式,分别给予按体质量和血压均衡分组的自发性高血压大鼠重组降血压肽-聚(乳酸-羟基乙酸)共聚物缓释微球,空白组给予等量生理盐水。结果口服给予rAHP缓释微球后8 h,自发性高血压大鼠的血压降至最低,且药效可以持续30 h;皮下注射重组降血压肽-聚(乳酸-羟基乙酸)共聚物缓释微球后20 h,血压降至最低,且可以持续约8 d。重组降血压肽-聚(乳酸-羟基乙酸)共聚物缓释微球对正常血压无影响。结论重组降血压肽-聚(乳酸-羟基乙酸)共聚物缓释微球具有显著的降压效果,并有明显的缓释作用。
Objective To investigate the antihypertensive effect of recombinant antihypertensive peptide (rAHP, sequence VLPVPR) on spontaneously hypertensive rats (SHR) with sustained release microspheres of biodegradable poly (lactic-co-glycolic acid) . Methods The oral administration and subcutaneous injection of two kinds of administration were given respectively according to the body mass and blood pressure balance of spontaneously hypertensive rats recombinant antihypertensive peptide - poly (lactic acid - glycolic acid) copolymer sustained release microspheres, the blank group given Equal amount of saline. Results The blood pressure of spontaneously hypertensive rats decreased to a minimum after 8 hours of oral administration of rAHP sustained-release microspheres, and the pharmacological effect could be sustained for 30 hours. The sustained release of recombinant hypotensive peptide-poly (lactic-co-glycolic acid) After 20 h of microspheres, blood pressure was reduced to a minimum and could last for about 8 days. Recombinant antihypertensive peptide - poly (lactic - co - glycolic acid) sustained - release microspheres had no effect on normotensive blood pressure. Conclusions The recombinant antihypertensive peptide-poly (lactic-co-glycolic acid) sustained-release microspheres has a significant antihypertensive effect, and has a significant sustained release effect.
