目的建立HPLC法测定雪莲种子提取物中牛蒡子苷含量的方法,为新疆雪莲特征性化合物的指认提供科学依据。方法采用Phenomenex Gemini色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-水梯度洗脱,流速1.0 ml/min,柱温30℃,检测波长270 nm。结果牛蒡子苷含量为0.24%且与其他成分分离良好,在105~1 050μg/ml范围内线性关系良好,r=0.999 7(n=6),对照品溶液低、中、高三种浓度的回收率分别为95.88%,102.33%,107.05%;RSD分别为3.88%,1.38%,1.02%。结论所建立的方法简便、准确、重现性好,并具有良好稳定性,为准确测定雪莲种子中牛蒡子苷含量提供可靠依据。
OBJECTIVE To establish a HPLC method for the determination of arctiin in Xuelian seed extract and provide a scientific basis for the identification of characteristic compounds in Xinjiang lotus root. Methods Phenomenex Gemini column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of a gradient of methanol and water. The flow rate was 1.0 ml / min and the column temperature was 30 ℃. The detection wavelength was set at 270 nm. Results The content of arctiin was 0.24% and was well separated from other components. There was a good linear relationship in the range of 105 ~ 1050μg / ml with r = 0.999 7 (n = 6). The recovery of low, medium and high concentrations of reference solution The rates were 95.88%, 102.33% and 107.05%, respectively. The RSDs were 3.88%, 1.38% and 1.02% respectively. Conclusion The established method is simple, accurate, reproducible and has good stability. It provides a reliable basis for the accurate determination of arctiin in the seeds of lotus seeds.
