目的:检测黄连含药血清对骨髓源巨噬细胞增殖的影响,探讨相应血清药理学实验中含药血清的合适浓度。方法:采集小鼠骨髓细胞诱导其单核细胞分化为巨噬细胞,流式细胞仪检测CD11b、F4/80检测细胞纯度;采用黄连水煎剂灌胃干预清洁级SD大鼠,常规制备含药血清,MTT法检测黄连含药血清对巨噬细胞增殖的影响。结果:与同浓度的正常血清对照组OD值比较,黄连含药血清在10%~40%浓度范围内对细胞增值无明显影响(P>0.05)。结论:在黄连含药血清干预巨噬细胞的相关实验中,含药血清加样浓度可选择在40%以内。
OBJECTIVE: To detect the effect of serum containing Coptis chinensis on the proliferation of bone marrow-derived macrophages, and to explore the appropriate concentration of serum containing drugs in the corresponding serum pharmacological experiments. Methods: Murine bone marrow cells were collected to induce monocytes to differentiate into macrophages. Flow cytometry was used to detect the purity of CD11b and F4 / 80 cells. The rats were treated with Coptis chinensis Serum and MTT assay of the serum of Coptis chinensis on the proliferation of macrophages. Results: Compared with the OD of normal control group at the same concentration, the drug-containing serum of Coptis chinensis had no significant effect on cell proliferation in the concentration range of 10% ~ 40% (P> 0.05). CONCLUSION: In the related experiments about the intervention of Coptis-containing serum on macrophages, the concentration of the medicated serum can be selected within 40%.
