目的 研究巨噬细胞的透明质酸 (HA)分布以及HA对于巨噬细胞黏附和迁移 ,探讨HA的作用机制。 方法 用聚集蛋白聚糖标记后 ,在光镜和共聚焦激光扫描显微镜下观察巨噬细胞内外的HA分布。 结果 巨噬细胞产生HA。静止细胞的HA多位于细胞表面、细胞膜下和核周。游走细胞内的HA主要分布在伪足、尾部和核周 ,细胞外的HA主要分布于伪足和尾部周围。巨噬细胞表面HA和底物HA增加细胞黏附 ,促进细胞迁移 ,但游离HA降低细胞黏附。 结论 HA在巨噬细胞的分布有特征性。细胞自身产生的HA和底物HA促进巨噬细胞黏附和迁移 ,而游离HA降低巨噬细胞的黏附。
Objective To study the distribution of hyaluronic acid (HA) in macrophages and the adhesion and migration of HA to macrophages, and to explore the mechanism of action of HA. Methods After labeling with aggrecan, the distribution of HA in macrophages was observed under light microscope and confocal laser scanning microscope. As a result macrophages produce HA. Quiescent cells are mostly located in the cell surface HA, sub-membrane and perinuclear. HA migrating within the cells mainly in the pseudopodia, caudal and perinuclear, extracellular HA mainly distributed in pseudopods and the tail around. Macrophage surface HA and substrate HA increase cell adhesion and promote cell migration, but free HA reduces cell adhesion. Conclusion The distribution of HA in macrophages is characteristic. The HA produced by the cells themselves and the substrate HA promote the adhesion and migration of macrophages, while the free HA reduces the adhesion of macrophages.
