目的检测SARS冠状病毒(SARS-CoV)S蛋白的功能性受体血管紧张素转换酶2(ACE2)在小鼠胰腺内、外分泌腺的表达。方法①RT-PCR:麻醉小鼠后取胰腺和肺组织,采用RT-PCR方法检测各组织ACE2的表达。②免疫电镜:麻醉小鼠后取胰尾,采用免疫电镜方法检测胰腺组织ACE2的表达。结果胰腺和肺脏组织均表达ACE2mRNA,且胰腺内、外分泌腺均表达ACE2。结论 ACE2在小鼠胰腺内、外分泌腺的表达可介导SARS-CoV侵入并损害胰腺,使胰岛素耗竭。
Objective To detect the expression of angiotensin-converting enzyme 2 (ACE2), a functional receptor of S protein of SARS coronavirus (SARS-CoV), in the pancreatic and exocrine glands of mice. Methods ①RT-PCR: The pancreas and lung tissue were taken after anesthetized mice. The expression of ACE2 in each tissue was detected by RT-PCR. ② Immunoelectron microscopy: After tailing the mice, the expression of ACE2 in pancreatic tissue was detected by immunoelectron microscopy. Results ACE2mRNA was expressed in both pancreas and lung tissues, and both endocrine and exocrine glands expressed ACE2. Conclusion The expression of exocrine gland of ACE2 in mouse pancreas can mediate the invasion of SARS-CoV and damage the pancreas, leading to the depletion of insulin.
