探讨脂多糖 (LPS)、硫酸鱼精蛋白及补肾活血泄浊汤对体外培养大鼠肾小球上皮细胞(GEC)蛋白质和硫酸肝素蛋白多糖 (HSPG)合成的影响。应用LPS、硫酸鱼精蛋白作为刺激因子 ,观察其对GEC蛋白质和HSPG合成的影响 ;并采用血清药理学方法 ,提取动物的含药血清作用于上述受刺激的上皮细胞 ,观察补肾活血泄浊汤对上皮细胞蛋白质和HSPG合成的影响。LPS和硫酸鱼精蛋白具有抑制GEC合成蛋白质及含硫化合物的作用 ;而补肾活血泄浊汤可显著拮抗上述因子的抑制细胞蛋白质和HSPG合成的作用。肾小球上皮细胞是补肾活血泄浊汤发挥治疗作用的重要靶细胞 ,这可能是该方防治以蛋白尿为主要临床表现的肾小球疾病的机制之一。
To investigate the effects of lipopolysaccharide (LPS), protamine sulfate, and Bushen Huoxue Xiezhuo Decoction on the synthesis of rat glomerular epithelial (GEC) protein and heparan sulfate proteoglycan (HSPG) in vitro. LPS and protamine sulphate were used as stimulators to observe its effect on the synthesis of GEC protein and HSPG; serum pharmacological methods were used to extract animal serum containing drugs acting on the above stimulated epithelial cells to observe Bushen Huoxue Xiezhuo Decoction. Effects on epithelial cell protein and HSPG synthesis. LPS and protamine sulfate have the effect of inhibiting GEC synthesis of protein and sulfur-containing compounds, while Bushen Huoxue Xiezhuo Decoction can significantly antagonize the inhibitory effects of these factors on cellular protein and HSPG synthesis. Glomerular epithelial cells are important target cells for the therapeutic effect of Bushen Huoxue Xiezhuo Decoction, which may be one of the mechanisms of the prevention and treatment of glomerular diseases with proteinuria as the main clinical manifestation.
