目的通过观察人类白细胞抗原G(HLA-G)在不同滋养细胞中的表达,探讨其与不明原因反复自然流产(RSA)的关系及免疫调节机制。方法用免疫酶细胞化学染色方法检测11例RSA患者滋养细胞的HLA-G表达情况,并以13例正常早孕的滋养细胞和JEG-3绒癌细胞作对照。结果RSA患者滋养细胞的HLA-G表达水平(1.15±1.56)显著低于正常早孕滋养细胞(8.97±2.76)和JEG-3绒癌细胞(9.56±1.81)(P﹤0.001)。后两种滋养细胞的HLA-G表达水平无显著性差异。RSA组5例表达为弱阳性,6例为阴性。正常早孕组8例表达为强阳性,5例为中等阳性。JEG-3绒癌细胞株表达为强阳性。结论滋养细胞的HLA-G表达与细胞浸润能力有关。RSA滋养细胞的低浸润能力可能与HLA-G的低表达有关。HLA-G表达可能不依赖母体环境,而在细胞内自身调节。
Objective To observe the expression of human leukocyte antigen G (HLA-G) in different trophoblasts and to explore its relationship with recurrent spontaneous abortion (RSA) of unexplained causes and its immunoregulatory mechanism. Methods The expression of HLA-G in gestational trophoblasts from 11 patients with RSA was detected by immunocytochemical staining, and 13 normal trophoblastic cells and JEG-3 choriocarcinoma cells were used as controls. Results The level of HLA-G expression in trophoblasts of RSA patients (1.15 ± 1.56) was significantly lower than that of normal pregnancy trophoblasts (8.97 ± 2.76) and JEG-3 choriocarcinoma cells (9.56 ± 1.81) (P <0.001). There was no significant difference in HLA-G expression level between the two trophoblasts. RSA group, 5 cases expressed weakly positive, 6 cases were negative. Normal early pregnancy group of 8 cases expressed strong positive, 5 cases were moderately positive. JEG-3 choriocarcinoma cell line expressed as strongly positive. Conclusion Trophoblast HLA-G expression and cell infiltration capacity. The low infiltration capacity of RSA trophoblast may be related to the low expression of HLA-G. HLA-G expression may not depend on the mother’s environment, but in the cell self-regulation.
