目的:观察槲皮素对人卵巢癌细胞系(3AO)增殖过程的影响。方法:以不同浓度槲皮素(1,10,20,40μmol/L)处理培养的3AO细胞后,采用活细胞计数法观察细胞增殖情况及去除槲皮素作用后细胞生长的变化;用氨基安替比林法测定碱性磷酸酶(ALP)活性。结果:槲皮素(1,10,20,40μmol/L)对3AO细胞的增殖有明显抑制作用,具有明显的剂量及时间依赖性;40μmol/L槲皮素作用5d后活细胞计数为对照的47.06%;去除其作用后,细胞生长得到恢复。用不同浓度(1,10,20,40μmol/L)的槲皮素处理3AO细胞5d后,ALP活性均有不同程度的减少,呈剂量依赖性;40μmol/L槲皮素作用3AO细胞不同时间,ALP活性随作用时间的延长而明显减少,呈时间依赖性。结论:槲皮素对3AO细胞的生长有明显的增殖抑制,去除其影响,细胞的生长可以部分恢复。
Objective: To observe the effect of quercetin on the proliferation of human ovarian cancer cell line (3AO). Methods: After treated with different concentrations of quercetin (1, 10, 20 and 40 μmol / L), the proliferation of 3AO cells was observed by the method of viable cell count and the cell growth was observed after treated with quercetin. Determination of alkaline phosphatase (ALP) activity by the method of probabilism. Results: Quercetin (1, 10, 20 and 40 μmol / L) significantly inhibited the proliferation of 3AO cells in a dose-and time-dependent manner. The viable cell counts of quercetin treated with 40 μmol / 47.06%. After removing its effect, cell growth was restored. After treated with quercetin at different concentrations (1, 10, 20 and 40μmol / L) for 5 days, the activity of ALP decreased in a dose-dependent manner. At 40μmol / L quercetin, ALP activity with the extension of time significantly reduced, in a time-dependent manner. CONCLUSION: Quercetin inhibits the proliferation of 3AO cells significantly, and the effect of quercetin on the growth of 3AO cells can be partially restored by removing the influence of quercetin.
