目的:探讨不同剂量的绿茶多酚对慢性脑低灌注大鼠抗氧化能力的影响。方法:健康雄性Wist-ar大鼠随机分为假手术组、2VO加生理盐水组、2VO加GTP100组(绿茶多酚100mg·kg-1·d-1)、2VO加GTP400组(绿茶多酚400mg·kg-1·d-1)。采用大鼠双侧颈总动脉结扎(2VO)制备慢性脑低灌注模型,结扎后从第4周开始给药,给药4周后采用生化方法检测脑皮质的抑制羟自由基能力和SOD的活力,并采用实时定量PCR方法检测皮质的Nrf2mRNA的表达。结果:绿茶多酚增强SOD活性和抑制羟自由基能力,增加皮质Nrf2mRNA的表达。400mg·kg-1·d-1剂量的绿茶多酚的作用更显著。结论:绿茶多酚增强慢性脑低灌注大鼠脑组织的抗氧化能力,尤其400mg·kg-1·d-1剂量的绿茶多酚作用更明显。
Objective: To investigate the effects of different doses of green tea polyphenols on anti-oxidation ability in rats with chronic cerebral hypoperfusion. METHODS: Healthy male Wist-ar rats were randomly divided into sham-operated group, 2VO plus normal saline group, 2VO plus GTP100 group (green tea polyphenol 100 mg·kg-1·d-1), 2VO plus GTP400 group (green tea polyphenol 400 mg Kg-1·d-1). Chronic cerebral hypoperfusion model was established by bilateral common carotid artery ligation (2VO) in rats. The administration was started from the fourth week after ligation. Biochemical methods were used to detect the inhibition of hydroxyl radical activity and SOD activity in cerebral cortex after 4 weeks of administration. The expression of Nrf2 mRNA in cortex was detected by real-time quantitative PCR. RESULTS: Green tea polyphenols enhanced SOD activity and inhibited hydroxyl radicals and increased cortical Nrf2 mRNA expression. The effect of 400 mg·kg-1·d-1 green tea polyphenols was more pronounced. CONCLUSION: Green tea polyphenols enhance the antioxidant capacity of brain tissue in rats with chronic cerebral hypoperfusion, especially the effect of 400 mg·kg-1·d-1 green tea polyphenols.
