目的:研究硝酸甘油对持续心肌缺血心肌细胞凋亡以及对凋亡相关基因的影响。方法:采用大鼠动物模型,用原位标记与半定量分析检测心肌细胞凋亡程度;以S-P免疫组化分别检测Fas基因蛋白。结果:持续心肌缺血组与假手术组相比,心肌细胞凋亡程度明显增加,Fas-L蛋白、Bcl-2蛋白的表达都有明显增高,差异有极显著性(P<0.01);硝酸甘油组与持续心肌缺血组比较,心肌细胞凋亡程度明显降低,差异有极显著性(P<0.01);Bcl-2蛋白表达增高,差异有显著性(P<0.05)、Fas-L蛋白表达降低,差异有显著性(P<0.05)。结论:硝酸甘油能够减轻持续心肌缺血过程中心肌细胞凋亡程度。
Objective: To study the effect of nitroglycerin on myocardial cell apoptosis and its related genes in myocardial ischemia. Methods: The rat myocardial cell model was used to detect the apoptosis of cardiomyocytes by using in situ labeling and semi-quantitative analysis. Fas gene protein was detected by S-P immunohistochemistry. Results: Compared with the sham-operated group, the apoptosis of cardiomyocytes in the continuous myocardial ischemia group was significantly increased, the expression of Fas-L protein and Bcl-2 protein were significantly increased, the difference was significant (P <0.01) Compared with continuous myocardial ischemia group, the degree of apoptosis of myocardial cells in glycerol group was significantly lower (P <0.01), the expression of Bcl-2 protein increased, the difference was significant (P <0.05), Fas-L protein The expression decreased, the difference was significant (P <0.05). Conclusion: Nitroglycerin can reduce the degree of myocardial cell apoptosis during myocardial ischemia.
