目的探讨桦褐孔菌多糖对高功率微波辐射致大鼠睾丸损伤的影响。方法 Wistar雄性大鼠30只,随机分为正常对照组、微波辐射模型组及治疗组、新微波辐射模型组及预防组,每组6只。除对照组外,其余组给予200mW/cm2微波辐照6min。对照组和模型组生理盐水灌胃,治疗组和预防组给予桦褐孔菌多糖2mL(400mg/kg)灌胃,每日1次,第11天处死各组大鼠,对各组大鼠精子密度、精子畸形率进行测定,以光镜和透射电镜观察睾丸的病理形态变化。结果短期高功率微波照射可引起大鼠精子密度减少,精子畸形率上升(P<0.05),同时睾丸发生明显的病理改变;而桦褐孔菌多糖灌胃后,治疗组和预防组与2个模型组比较,精子密度增加,精子畸形率下降(P<0.05),光镜下生精细胞和间质细胞损伤以及血管扩张充血情况都明显减轻,超微结构显示线粒体肿胀和内质网扩张也明显减轻,其中治疗组各级生精细胞损伤和炎症反应均比预防组轻。结论桦褐孔菌多糖对微波辐射致睾丸损伤具有显著防护效果,治疗性给药的效果较预防性给药更加显著。
Objective To investigate the effects of Inonotus obliquus polysaccharides on testicular injury induced by high power microwave in rats. Methods Thirty male Wistar rats were randomly divided into normal control group, microwave radiation model group and treatment group, new microwave radiation model group and preventive group, with 6 rats in each group. Except the control group, the other groups were given microwave irradiation of 200mW / cm2 for 6min. The rats in the control group and the model group were given gavage orally. The treatment group and prophylaxis group were given 2mL (400mg / kg) of Inonotus obliquus polysaccharide to the rats, once a day, the rats in each group were killed on the 11th day, Density, sperm deformity rate were measured to observe the pathological changes of testes with light and transmission electron microscopy. Results Short-term high-power microwave irradiation caused a decrease in sperm density and an increase in sperm deformity in rats (P <0.05), and a marked pathological change was observed in testis. After administration of polysaccharides of Inonotus distichodon, Compared with the model group, the sperm density increased and the sperm deformity rate decreased (P <0.05). The damage of spermatogenic cells and interstitial cells under light microscopy and vasodilation and congestion were significantly reduced. The ultrastructure showed mitochondrial swelling and endoplasmic reticulum dilatation Significantly reduced, of which the treatment group at all levels of spermatogenic cell damage and inflammatory response were lighter than the prevention group. Conclusions The polysaccharide from Inonotus obliquus has a significant protective effect on the testicular damage induced by microwave radiation. The therapeutic effect of the therapeutic administration is more significant than that of prophylactic administration.
