目的:探讨冬凌草植株再生过程中培养物的生理特性与冬凌草甲素积累之间的机制。方法:采用紫外分光光度法、高效液相法测定不同时期培养物内叶绿素、可溶性糖、可溶性蛋白、丙酮酸、冬凌草甲素的含量。结果:培养物内叶绿素、丙酮酸、可溶性蛋白含量与冬凌草甲素含量呈正相关,相关系数分别为0.781,0.897,0.866;而可溶性糖含量与冬凌草甲素含量呈负相关,相关系数为-0.515。结论:冬凌草培养物内叶绿素、丙酮酸的含量、可溶性蛋白与冬凌草甲素积累有相互促进的关系,而可溶性糖变化主要与培养物细胞的生长、发育关系密切。
Objective: To explore the mechanism of the physiological characteristics of the culture and the accumulation of Rubescensine A in the process of plant regeneration from Rabdosia rubescens. METHODS: The contents of chlorophyll, soluble sugar, soluble protein, pyruvic acid, oridonin were determined by UV spectrophotometry and high performance liquid chromatography. Results: The contents of chlorophyll, pyruvic acid and soluble protein in the culture were positively correlated with the content of Rubescensine A. The correlation coefficients were 0.781, 0.897, and 0.866, respectively. The soluble sugar content was negatively correlated with Rubescensine A content. It is -0.515. Conclusion: The contents of chlorophyll and pyruvic acid and the accumulation of soluble protein and Rubescensine A in the culture of Rabdosia rubescens have a mutual promotion relationship, while the changes of soluble sugar are mainly related to the growth and development of culture cells.
