目的研究康复训练对大鼠脑梗死灶周围微管蛋白(tubulin)表达的影响。方法40只Wistar大鼠,采用Longa颈外动脉线栓法制备大鼠大脑中动脉闭塞模型,造模后将大鼠随机分为康复组和造模对照组,每组20只。康复组大鼠每天进行滚筒、平衡木、转棒及网屏训练,时间1h;造模对照组置于普通笼中饲养,不予以康复训练。每组分别于造模后3,7,14和21d4个时间点处死5只大鼠,并取脑组织,采用免疫组织化学方法观察每个时间点脑梗死灶周围tubulin的表达。结果康复组神经功能评分明显低于造模对照组(P<0.05)。2组tubulin阳性细胞于脑缺血3d后即已出现,7~21d有大量表达,14d达高峰;康复组于脑缺血7d和21d,tubulin阳性细胞的表达水平明显高于造模对照组(P<0.05)。结论康复训练能促使中枢神经系统表达较高水平的tubulin。
Objective To study the effect of rehabilitation training on the expression of tubulin in the cerebral infarction of rats. Methods Forty Wistar rats were randomly divided into rehabilitation group and model control group, 20 rats in each group. Rats in rehabilitation group were trained on drum, balance beam, rotating rod and screen every day for 1 hour. The model control group was housed in ordinary cage and was not rehabilitated. Five rats in each group were sacrificed at 3, 7, 14 and 21 days after modeling respectively. The brain tissues were removed and the expression of tubulin around cerebral infarction at each time point was observed by immunohistochemistry. Results The neurological function score of rehabilitation group was significantly lower than that of the model control group (P <0.05). The tubulin positive cells in group 2 appeared three days after cerebral ischemia, and reached the peak on day 7-21, reaching the peak on day 14. The expression of tubulin positive cells in the rehabilitation group was significantly higher than that in the model control group at 7d and 21d P <0.05). Conclusion Rehabilitation training can promote the expression of tubulin in the central nervous system at a higher level.
