目的:探讨健胃愈疡颗粒对大鼠胃溃疡模型溃疡愈合及胃组织IL-1β蛋白表达的影响。方法:用Okabe改良法复制SD大鼠胃溃疡模型,观察溃疡指数,采用免疫组织化学法观察胃粘膜组织形态及IL-1β蛋白表达的变化。结果:①健胃愈疡组的溃疡指数明显低于模型组和法莫替丁组;②HE染色法显示健胃愈疡组较模型组及法莫替丁组黏膜厚度增加,溃疡床及溃疡周围炎性细胞浸润明显减少,纤维排列较为整齐。③IL-1β在正常胃组织呈阴性或偶有阳性信号表达,造模型7 d后表达显著增加,健胃愈疡组和法莫替丁组低于模型组(P<0.01);14 d后各组的阳性信号表达均降至接近正常。结论:健胃愈疡颗粒能保护大鼠胃粘膜组织,降低胃溃疡大鼠模型溃疡指数,改善溃疡愈合质量,减少胃组织IL-1β蛋白的表达,促进溃疡愈合。
Objective: To investigate the effect of Jianweiyuyang granule on ulcer healing and expression of IL-1β protein in gastric tissue in gastric ulcer model rats. METHODS: SD rat gastric ulcer model was modified by Okabe’s modified method, and the ulcer index was observed. The changes of gastric mucosal tissue morphology and IL-1β protein expression were observed by immunohistochemistry. Results: 1 The ulcer index of Jianweiyuyang group was significantly lower than that of model group and famotidine group; 2HE staining showed that the mucosal thickness of Jianweiyuyang group was higher than that of model group and famotidine group, ulcer bed and ulcer circumference. The infiltration of inflammatory cells was significantly reduced and the fibers were arranged more neatly. 3The expression of IL-1β was negative or occasionally positive in normal gastric tissues. The expression of IL-1β was significantly increased at 7 days after model establishment, while that of Jianweiyuyang and famotidine was lower than that of model group (P<0.01). The positive signal expression in the group dropped to near normal. Conclusion: Jianweiyuyang granule can protect rat gastric mucosal tissue, reduce the ulcer index of gastric ulcer rat model, improve the quality of ulcer healing, reduce the expression of gastric tissue IL-1β protein and promote ulcer healing.
