目的研究人用皮内注射布氏菌活疫苗拟取代目前我国人用皮上划痕布氏菌活疫苗。方法对菌种进行了全面检定合格后,进行菌苗制备及实验室动物毒理、药效学方面的系列研究包括小白鼠急性毒性试验、局部刺激试验、全身过敏试验、长期毒性试验、用细胞免疫诊断制剂布氏菌素对皮内免疫12周的豚鼠做背部皮肤变态反应试验。结果小鼠皮内注射布氏菌活疫苗的最大耐受量为10亿个菌,用布氏菌素注射足底,可引起明显迟发型变态反应。局部刺激试验只有注射10亿菌的部位会在48h后化脓,但在1~2周内可自愈。全身过敏试验,皮内免疫布氏菌疫苗组及阴性对照组对豚鼠进行攻击后,均未出现任何异常反应,而阳性对照组豚鼠全部死亡。毒性试验,布氏活疫苗皮内免疫组与盐水对照组经6周和12周后各项血常规、血生化检查均未见异常,各脏器组织的病理学检查亦未见病理改变,免疫12周后对豚鼠骨髓造血细胞亦无影响,对皮内免疫12周的豚鼠做背部皮肤变态反应试验,24~48h结果为阳性。结论动物实验结果显示,布氏菌活疫苗用皮内注射的途径进行免疫是可行的。
Objective To study human intradermal injection of live Brucella bacteria vaccine to replace the current human scratches on the skin of Brucella live vaccine. Results The maximum tolerated dose of intradermal injection of live Brucella vaccine in mice was 1 billion bacteria. Injecting the plantarum with brucellin could cause obvious delayed-type hypersensitivity. Local irritation test only parts of the injection of 1 billion bacteria purulent after 48h, but in 1 to 2 weeks can be self-healing. Systemic allergy test, intradermal immune Brucella vaccine group and negative control group of guinea pigs after attack, did not appear any abnormal reaction, and the guinea pigs of the positive control group all died. Toxicity tests, intradermal bursal vaccine and saline control group after 6 weeks and 12 weeks of blood tests, blood biochemical tests were no abnormalities, pathological examination of the organs of the organization also showed no pathological changes, immune Twelve weeks later had no effect on the hematopoietic cells of guinea pig bone marrow. The guinea pigs on the 12th week of intradermal immunization were tested for back skin allergy. The result of 24 ~ 48h was positive. Conclusion The results of animal experiments show that viable intradermal injection of live Brucella vaccine is feasible.
