目的:对糙海参进行质量研究,初步建立糙海参的质量标准。方法:利用显微法鉴别骨片,显色法鉴别己糖醛酸和酸性多糖;采用1,9-二甲基亚甲蓝显色分光光度法测定酸性多糖的含量,其余检查项按照《中国药典》2010年版一部进行检测。结果:糙海参骨片有扣状体,少量桌形体、穿孔板和杆状体;糙海参体壁中含有己糖醛酸和酸性多糖;酸性多糖含量不低于2.0%,其含量测定方法在0.04~0.18 mg·mL-1范围内呈现良好的线性关系,平均回收率为94.51%(RSD4.4%),精密度RSD0.58%;杂质含量限度不超过15.0%,水分含量限度10.0%,总灰分和酸不溶性灰分含量限度60.0%和20.0%。结论:对糙海参的来源、性状、鉴别、检查、含量测定等进行了研究,为糙海参质量标准的建立、规范和完善提供科学依据。
Objective: To study the quality of sea cucumber and establish the quality standard of sea cucumber. Methods: The bone chips were identified by microscopy, and the hexuronic acid and acid polysaccharides were identified by colorimetric method. The content of acid polysaccharides was determined by 1, 9-dimethylmethane blue colorimetric spectrophotometry. Pharmacopoeia "2010 edition of a test. Results: The crude sea cucumber bone chips have a buckle-like body, a small amount of table-shaped body, perforated plate and rod-shaped body; rough sea cucumber body wall containing hexuronic acid and acid polysaccharides; acid polysaccharide content of not less than 2.0% The average recovery was 94.51% (RSD4.4%) and the precision RSD was 0.58%. The impurity content limit was less than 15.0% and the moisture content limit was 10.0% Total and acid-insoluble ash content limits of 60.0% and 20.0%. CONCLUSIONS: The origin, traits, identification, examination and determination of the content of rough sea cucumber were studied in order to provide a scientific basis for the establishment, standardization and perfection of the quality standard of sea cucumber.
