目的:探讨肝癌患者外周血淋巴细胞染色体脆性部位与原癌基因表达率的相关性。方法:采用低叶酸、低小牛血清、较高pH值和G显带培养方法,常规外周血染色体制片。对20例住院肝癌患者(实验组)和18例献血队员(对照组)的外周血淋巴细胞染色体脆性部位(Fra)及染色体畸变(CAR)进行分析。结果:实验组检出44种脆性部位,主要分布于A、B、C组染色体,其表达率为9.90%,与对照组(0.67%)比较,差异有显著性(P<0.01)。脆性部位中有11种与原癌基因重叠,重叠率为25%。结论:检出染色体的脆性部位可能与肝癌的发生、发展有关。
Objective: To investigate the correlation between the chromosome fragile sites and the expression rates of proto-oncogenes in peripheral blood lymphocytes from patients with hepatocellular carcinoma. Methods: Low-folate, low-calf serum, higher pH and G-banding culture methods, and conventional peripheral blood chromosome production. The Fragile Fragments (Fr) and Chromosomal Aberrations (CAR) of peripheral blood lymphocytes from 20 hospitalized liver cancer patients (experimental group) and 18 blood donors (control group) were analyzed. Results: 44 fragile sites were detected in the experimental group, which were mainly distributed in the chromosomes of group A, B, and C, and the expression rate was 9.90%. Compared with the control group (0.67%), the difference was statistically significant (P<0.01). Eleven of the fragile sites overlap with the proto-oncogene, with an overlap of 25%. Conclusion: Detecting the brittle parts of chromosomes may be related to the occurrence and development of liver cancer.
