目的:研究双脱甲氧基姜黄素体外抑制SKOV3卵巢癌细胞抑制作用的体外实验研究。方法:(1)将不同浓度双脱甲氧基姜黄素(0、10ug/ml、20ug/ml、40ug/ml)作用于人卵巢癌SKOV3细胞6、12、24h,采用四甲基偶氮唑盐(MTT)比色法检测SKOV3细胞的增殖活性;(2)流式细胞仪观察双脱甲氧基对SKOV3卵巢癌细胞周期的作用,免疫组化检测增殖细胞核抗原(PCNA)的表达。结果:(1)双脱甲氧基姜黄素对SKOV3细胞具有增殖抑制作用,且具有浓度和时间依赖效应;(2)双脱甲氧基姜黄素能诱导SKOV3细胞G1期阻滞,通过抑制细胞生长周期,降低细胞增生速率抑制肿瘤细胞增殖;(3)双脱甲氧基姜黄素能下调SKOV3细胞PCNA蛋白表达,并呈一定的浓度依赖性。结论:双脱甲氧基姜黄素能明显抑制人卵巢癌SKOV3细胞的体外增殖,可能为临床治疗卵巢癌提供了一种新的方法。
Objective: To study the in vitro study of the inhibitory effect of bisdemethoxy curcumin on SKOV3 ovarian cancer cells in vitro. Methods: (1) Human ovarian cancer SKOV3 cells were treated with different concentrations of bisdemethoxy curcumin (0, 10ug / ml, 20ug / ml and 40ug / ml) (MTT) assay was used to detect the proliferation activity of SKOV3 cells. (2) Flow cytometry was used to observe the effects of dideoxymethylation on the cell cycle of SKOV3 ovarian cancer cells. The expression of proliferating cell nuclear antigen (PCNA) was detected by immunohistochemistry. RESULTS: (1) Didemethoxy curcumin inhibited the proliferation of SKOV3 cells in a concentration-and time-dependent manner. (2) Dideoxy-curcumin induced G1 arrest of SKOV3 cells, Growth cycle, reduce the rate of cell proliferation and inhibit tumor cell proliferation; (3) didemethoxy curcumin can down-regulate the expression of PCNA protein in SKOV3 cells in a concentration-dependent manner. CONCLUSION: Demethoxycurcumin can significantly inhibit the proliferation of human ovarian cancer cell line SKOV3 in vitro and may provide a new method for clinical treatment of ovarian cancer.
