目的研究原癌基因人表皮生长因子受体2(HER2)对MCF10A人乳腺上皮细胞形态和迁移能力的影响及机制。方法通过慢病毒包装及感染MCF10A人乳腺上皮细胞,建立HER2过表达细胞,用TranswellTM法研究细胞迁移能力变化,结合Western blot法、实时定量PCR和免疫荧光染色检测细胞中上皮钙黏素(E-cadherin)的表达。结果过表达HER2的乳腺上皮细胞呈分散型生长,细胞迁移能力增强,E-cadherin蛋白表达受到抑制,细胞膜上E-cadherin表达也显著降低,重新过表达E-cadherin能抑制HER2诱导的细胞迁移和分散。结论 HER2能通过抑制E-cadherin促进人乳腺上皮细胞迁移。
Objective To investigate the effect and mechanism of the oncogene human epidermal growth factor receptor 2 (HER2) on the morphology and migration of MCF10A human mammary epithelial cells. METHODS: HER2 overexpression cells were established by lentivirus packaging and infection of MCF10A human breast epithelial cells. TranswellTM was used to study the changes of cell migration. Western blot, real-time quantitative PCR and immunofluorescence staining were used to detect the expression of E- cadherin expression. RESULTS: HER2-overexpressing mammary epithelial cells showed decentralized growth, increased cell migration, inhibition of E-cadherin protein expression, and decreased expression of E-cadherin on the cell membrane. Overexpression of E-cadherin inhibited HER2-induced cell migration and dispersion. Conclusion HER2 can promote human mammary epithelial cell migration by inhibiting E-cadherin.
