肝癌的浸润和转移的过程中,基底膜(basement membrane,BM)必将降解.BM 主要由Ⅳ型胶原、糖蛋白等组成,呈厚实的网状结构,而Ⅳ型胶原又是 BM 的主要支架.许多资料研究表明,肿瘤细胞分泌的基质金属蛋白酶(matrix metalloproteinases,MMPs)中的 MMP-2是最为主要的基质金属蛋白酶,其降解底物是Ⅳ胶原,使 BM 发生缺损,促使肿瘤细胞向周围组织浸润和转移.MMP-2以前酶(Pro-MMP-2)形式存在,需经蛋白酶等水解和修饰,才具有生物学活性.为此,我们利用酶谱(zymography)和 UVP 软件检测并分析了32例肝细胞癌中MMP-2水平及活性比,旨在探讨其与肝细胞癌浸润、转移的关系.
