目的:比较大鼠脊髓单胺类神经递质的含量和基础释放量。方法:以反相离子高效液相色谱电化学检测法对样品中的单胺类递质及其代谢产物进行定量分析。结果:(1)分别测得了大鼠脑脊液、脊髓灌流液和脊髓背部组织提取液中单胺类递质及其代谢产物的含量;(2)在大鼠脊髓背部组织中同时含有5羟色胺、去甲肾上腺素和多巴胺,但在脊髓灌流液和脑脊液中只能检测到去甲肾上腺素,部分样品中能测到多巴胺;(3)递质代谢产物在脑脊液和脊髓灌流液中的含量均高于其对应的递质。结论:该方法的分辨率和灵敏度均能满足实验要求,所得数据为进一步研究脊髓水平单胺类递质的代谢和生理功能奠定了基础。
OBJECTIVE: To compare monoamine neurotransmitters and basal release of spinal cord in rats. Methods: Quantitative analysis of monoamine neurotransmitters and their metabolites in samples was carried out by reversed-phase high performance liquid chromatography with electrochemical detection. Results: (1) The contents of monoamine neurotransmitters and their metabolites in the cerebrospinal fluid, spinal perfusate and spinal cord tissue of the spinal cord were measured respectively. (2) Serotonin, Norepinephrine and dopamine, but only norepinephrine was detected in spinal perfusate and cerebrospinal fluid, and dopamine was detected in some samples. (3) The contents of neurotransmitter metabolites in cerebrospinal fluid and spinal cord perfusate were both high In its corresponding transmitter. Conclusion: The resolution and sensitivity of the method can meet the experimental requirements, the data obtained for the further study of spinal cord monoamine neurotransmitter metabolism and physiological function laid the foundation.
