目的:探讨舒肝健脑调郁片的抗抑郁机制,为临床应用提供依据。方法:采用大鼠抑郁动物模型为慢性轻度不可预见性的应激加单居模型,造模成功后随机分为5组,每组12只,氟西汀组0.75mg·kg-1·d-1、舒肝健脑调郁片低剂量组1.8g·kg-1·d-1、舒肝健脑调郁片高剂量组3.6g·kg-1·d-1,正常组和模型组1ml·kg-1·d-1生理盐水灌胃,从造模第1d开始,1次/d。至动物处死当天共21d。采用透射电镜观察细胞超微结构变化和免疫组化法检测C-FOS和C-JUN变化。结果:舒肝健脑调郁片能够减轻应激造成的海马神经元损伤;C-FOS和C-JUN蛋白免疫反应阳性细胞平均目标灰度值高剂量组与模型组比较显著增高。结论:表明舒肝健脑调郁片能够减轻应激造成的海马神经元损伤,对慢性应激状态下导致的抑郁症有治疗作用。
Objective: To investigate the anti-depression mechanism of Shugan Jiannao Tiaoyu Tablets and provide basis for clinical application. METHODS: The animal model of depression in rats was used as a chronic mild unpredictable stress plus single-living model. After successful modeling, the rats were randomly divided into 5 groups with 12 in each group. Fluoxetine group was 0.75 mg·kg-1·d. - 1, Shugan Jiannao Tiaoyu tablets low dose group 1.8g · kg-1 · d-1, Shuganjiannao Tiaoyu tablets high dose group 3.6g · kg-1 · d-1, normal group and model group 1 ml·kg-1·d-1 saline was intragastrically administered, once from the first day of modeling, once a day. A total of 21 days had elapsed since the animals were executed. Transmission electron microscopy was used to observe the ultrastructure changes of cells and immunohistochemical method to detect the changes of C-FOS and C-JUN. Results: Shugan Jiannao Tiaoyu Tablet could relieve the damage of hippocampal neurons induced by stress. The mean target gray value of C-FOS and C-JUN protein immunoreactive positive cells was significantly higher in the high-dose group than in the model group. Conclusion: Shugan Jiannao Tiaoyu Tablets can reduce the damage of hippocampal neurons caused by stress and have therapeutic effects on depression caused by chronic stress.
