目的探讨50 mg/L表没食子儿茶素-3-没食子酸酯(EGCG)对小鼠皮肤外伤愈合组织基质金属蛋白酶1(MMP-1)mRNA表达及MMP-1水平的影响。方法将57只小鼠随机分为对照组(含无损伤组和损伤对照组)、50 mg/L EGCG处理组和辅料组,除无损伤组为3只外,其余各组均为18只,然后分别于损伤后6、12、24、72、120、168 h取损伤愈合皮肤。利用RT-PCR和酶联免疫吸附法(ELISA)检测皮肤外伤愈合期间损伤愈合组织MMP-1 mRNA表达水平和MMP-1含量。结果检测结果表明,50 mg/L EGCG能在皮肤外伤愈合晚期(72 h后)抑制损伤愈合组织MMP-1 mRNA表达,降低MMP-1含量。结论 EGCG对皮肤外伤愈合、晚期愈合组织MMP-1mRNA表达和MMP-1生成具有抑制作用,提示50 mg/L的EGCG能促进皮肤外伤愈合。
Objective To investigate the effects of epigallocatechin-3-gallate (EGCG) 50 mg / L on the expression of matrix metalloproteinase 1 (MMP-1) mRNA and the level of MMP-1 in skin wound healing mice. Methods Fifty-seven mice were randomly divided into control group (non-injury group and injury control group), 50 mg / L EGCG treatment group and excipient group, except no injury group was 3, the other groups were 18, Then, 6, 12, 24, 72, 120, 168 h after injury were taken to heal the healing skin. RT-PCR and enzyme-linked immunosorbent assay (ELISA) were used to detect the expression of MMP-1 mRNA and MMP-1 in wound healing tissue during wound healing. Results The results showed that 50 mg / L EGCG could inhibit the expression of MMP-1 mRNA and decrease the content of MMP-1 in the wound healing tissues at the later stage of skin wound healing (after 72 h). Conclusion EGCG can inhibit skin wound healing, MMP-1 mRNA expression and MMP-1 production in advanced healing tissues, suggesting that 50 mg / L EGCG can promote skin wound healing.
