目的:研究难治性癫患者脑组织内钙黏蛋白-18(cadherin-18)mRNA的表达,拟从另一角度加深对人类难治性癫的认识。方法:在基因芯片扫描的基础上,运用RT-PCR检测钙黏蛋白-18mRNA在难治性癫患者脑部的变化。结果:基因芯片扫描显示钙黏蛋白-18mRNA在难治性癫患者脑部比正常人表达上调5.320倍,RT-PCR检测同样显示钙黏蛋白-18mRNA在难治性癫患者脑部表达增强,其电泳条带灰度值为25.547,而正常组仅为11.176。结论:结果表明特异性表达于中枢神经系统的钙黏蛋白-18mRNA在难治性癫患者脑部表达增强,提示钙黏蛋白-18可能参与了人类难治性癫的形成。
Objective: To study the expression of cadherin-18 mRNA in the brain tissue of patients with intractable epilepsy, and to deepen the understanding of intractable epilepsy in another aspect. Methods: The changes of cadherin-18 mRNA in the brain of refractory epilepsy patients were detected by RT-PCR on the basis of gene chip scanning. Results: Gene microarray showed that cadherin-18 mRNA was up-regulated by 5.320-fold in brains of patients with intractable epilepsy compared with normal controls. RT-PCR also showed that cadherin-18 mRNA was increased in the brains of patients with intractable epilepsy The electrophoresis band gray value was 25.547, while the normal group was only 11.176. Conclusions: The results show that cadherin-18 mRNA specifically expressed in the central nervous system is enhanced in the brain of refractory epilepsy patients, suggesting that cadherin-18 may be involved in the formation of human refractory epilepsy.
