目的:研究长梗秦艽酮抑制人肝癌BEL-7402细胞生长与Akt和ERK1/2通路的关系。方法:采用MTT法观察BEL-7402细胞活性,Western blot蛋白质印记法检测Akt和ERK1/2磷酸化水平,流式细胞术分析细胞周期。结果:长梗秦艽酮能够抑制人肝癌BEL-7402细胞生长,诱导S期细胞周期阻滞,激活Akt和ERK1/2磷酸化,并且Akt和ERK1/2抑制剂能够拮抗长梗秦艽酮对BEL-7402细胞的生长抑制和S期阻滞诱导作用。结论:长梗秦艽酮可以通过调节Akt和ERK1/2通路诱导肿瘤细胞周期阻滞,从而抑制肿瘤细胞生长。
OBJECTIVE: To study the relationship between the long-stemmed genistein inhibiting the growth of human hepatocellular carcinoma BEL-7402 cells and Akt and ERK1 / 2 pathways. Methods: The activity of BEL-7402 cells was observed by MTT assay. The phosphorylation levels of Akt and ERK1 / 2 were detected by Western blot. The cell cycle was analyzed by flow cytometry. Results: Long Qin Qin ketones can inhibit the growth of human hepatocellular carcinoma BEL-7402 cells, induce cell cycle arrest in S phase, activate Akt and ERK1 / 2 phosphorylation, and Akt and ERK1 / 2 inhibitors can antagonize long- 7402 cell growth inhibition and induction of S phase arrest. Conclusion: Long Qin Qin ketone can induce tumor cell cycle arrest by regulating Akt and ERK1 / 2 pathways, thereby inhibiting tumor cell growth.
