目的建立测定板栗种仁酸水解前后原儿茶酸含量的方法。方法采用HPLC法,色谱柱为Diamonsil C18柱(200 mm×4.6 mm,5μm),流动相为甲醇-水-磷酸(体积比为8.0∶92.0∶0.1),检测波长为259 nm,流速为0.8 mL.min-1,柱温为25℃。结果测定酸水解前板栗种仁中原儿茶酸含量质量分数为8.12×10-3%;酸水解后原儿茶酸含量质量分数为1.82×10-2%。结论HPLC法可用于板栗种仁中原儿茶酸的含量测定;板栗种仁提取物经过酸水解后的原儿茶酸含量有较大增加。
Objective To establish a method for determining the content of protocatechuic acid before and after hydrolysis of castor seed kernel acid. Methods HPLC method was used. The chromatographic column was Diamonsil C18 column (200 mm×4.6 mm, 5 μm). The mobile phase consisted of methanol-water-phosphoric acid (volume ratio of 8.0:92.0:0.1). The detection wavelength was 259 nm and the flow rate was 0.8 mL. .min-1, column temperature 25°C. Results The content of protocatechuic acid in chestnut kernels before acid hydrolysis was 8.12×10-3%, and the acid content in protocatechuic acid after acid hydrolysis was 1.82×10-2%. Conclusion The HPLC method can be used to determine the content of protocatechuic acid in chestnut seeds. The content of protocatechuic acid in the chestnut seed kernels after acid hydrolysis is greatly increased.
