目的:观察性激素对育龄期和绝经期妇女不同部位脂肪组织11β羟基类固醇脱氢酶Ⅰ型(11β-hydroxysteroid dehy-drogenase type 1,11βHSD1)mRNA表达水平的调控作用。方法:育龄期非妊娠妇女6例,年龄(29.83±3.43)岁及绝经期妇女16例,年龄(65.19±6.81)岁,取其皮下和网膜脂肪组织进行培养,应用雌激素和雄激素刺激脂肪组织24h,实时定量PCR方法测定脂肪组织11βHSD1 mRNA水平。结果:雌激素可使绝经期妇女皮下脂肪组织11βHSD1 mRNA表达明显增加(P<0.05),使网膜脂肪组织11βHSD1 mRNA表达水平下降30%。雄激素刺激可抑制育龄期妇女皮下脂肪组织11βHSD1 mRNA表达近40%(P=0.001),而使绝经期妇女网膜脂肪组织11βHSD1 mRNA表达增加1.9倍(P=0.005)。结论 :性激素对脂肪组织11βHSD1 mRNA表达的调控呈现年龄和部位特异性改变,可能是不同时期女性体脂分布的重要分子生物学机制。
Objective: To observe the effect of sex hormones on the mRNA expression of 11β-hydroxysteroid dehy-drogenase type 1, 11βHSD1 in different parts of women of childbearing age and menopause. Methods: Six non-pregnant women of childbearing age, 29.83 ± 3.43 years of age and 16 cases of menopausal women aged 65.19 ± 6.81 years old, were subcutaneously and omental adipose tissue culture, application of estrogen and androgen to stimulate fat Tissue 24h, real-time quantitative PCR method for determination of adipose tissue 11βHSD1 mRNA levels. Results: Estrogen significantly increased the expression of 11βHSD1 mRNA in subcutaneous adipose tissue of postmenopausal women (P <0.05), and decreased the expression level of 11βHSD1 mRNA in omental adipose tissue by 30%. Androgen stimulation inhibited the expression of 11βHSD1 mRNA in subcutaneous adipose tissue of women of childbearing age by nearly 40% (P = 0.001), while the mRNA expression of 11βHSD1 in omental adipose tissue of menopausal women increased 1.9 times (P = 0.005). Conclusion: The regulation of sex hormone on the expression of 11βHSD1 mRNA in adipose tissue shows age-specific and site-specific changes and may be an important molecular biological mechanism of body fat distribution in different periods.
