目的:研究通络驻景丸对糖尿病视网膜病变代谢记忆机制中炎症因子IL-6、TNF-a、hs CPR及视网膜NF-κB表达的影响。方法:链脲佐菌素(STZ)诱导SD大鼠糖尿病,造模成功的大鼠分成6组:模型组(B组)、代谢记忆对照组(C组)、阳性对照组(D组)和通络驻景丸大中小剂量干预组(E、F、G组),另设一组不造模的空白对照组(A组)。A组、B组、C组给予蒸馏水,D组给予阳性药,E、F、G组分别给通络驻景丸大、中、小剂量。每日灌胃给药一次,容积1ml/100g体质量,连续6个月。从第四个月开始除A、B组外其余5组给予胰岛素控制血糖至正常。实验期间每周检测血糖一次,6月后检测各组血清中炎性因子及NF-κB在视网膜上的表达,比较炎性因子水平及NF-κB在视网膜上的表达。结果:与C组比较,阳性药可降低模型动物血清中IL-6水平;通络驻景丸大、中剂量可降低模型动物血清中IL-6、TNF-a、hs CPR水平,通络驻景丸大剂量可抑制视网膜NF-κB高表达;B组与C组比较无统计学意义。结论:糖尿病后期即使血糖控制良好,炎性因子的水平也是高表达;通络驻景丸可降低糖尿病视网膜病变代谢记忆机制中炎性因子的水平及视网膜上NF-κB的高表达。
Objective: To investigate the effects of Tongluojingkangwan on the expression of inflammatory cytokines IL-6, TNF-a, hs CPR and retinal NF-κB in the metabolic memory mechanism of diabetic retinopathy. Methods: STZ-induced diabetic rats were divided into 6 groups: model group (B group), metabolic memory control group (C group), positive control group (D group) and model group Tongluo Jingwang Dazhong Dansu intervention group (E, F, G), another set of non-model control group (A). Group A, B, C were given distilled water, D group were given positive drug, E, F, G groups were given Tongluo Jing Jue Wan large, medium and small doses. Gavage once a day, volume 1ml / 100g body weight for 6 months. From the fourth month except the A and B groups, the remaining five groups were given insulin to control blood sugar to normal. The blood glucose was measured once a week during the experiment. The levels of inflammatory cytokines and NF-κB in the retina were detected after 6 months. The levels of inflammatory cytokines and the expression of NF-κB in the retina were also compared. Results: Compared with group C, the positive drug can reduce the level of IL-6 in the serum of model animals. Tonglujinguwan large and medium dose can reduce the levels of IL-6, TNF-a and hs CPR in model animal serum, Large doses of pills can inhibit retinal NF-κB high expression; B group and C group was not statistically significant. CONCLUSION: Even if blood glucose is well controlled during the later stage of diabetes, the level of inflammatory cytokines is also high. Tongluojijing pill can reduce the level of inflammatory cytokines and the high expression of NF-κB on the retina in diabetic retinopathy.
