目的:探讨炎性细胞因子IL-1α、L-1β、IL-6和TNF-α在尿结石形成中的作用,以及α-亚麻酸(α-LNA)对其的抑制影响。方法:将45只雄性Wistar成年大鼠随机分成3组,分别接受普通饲料加自来水饮水(A组)、普通饲料加诱石剂饮水(B组)和普通饲料加诱石剂饮水并每天用苏子油灌胃(2 g.只-1.d-1)(C组)。于喂养第8周后检测各组大鼠血细胞因子水平。利用体外培养的大鼠肾系膜细胞(RMC),设立空白对照(a组)、IL-6处理(b组)、EPA10μmol加IL-6处理(c组)、EPA50μmol加IL-6处理(d组)及无水乙醇处理(e组),用放免法检测培养上清中PGE2含量。结果:C组和A组IL-1αI、L-6水平显著低于B组,三组间IL-1β、TNF-α水平差异无统计学意义。细胞培养上清PGE2含量,a、c、d组显著低于b组,d组显著低于c组,a、e两组间差异亦无统计学意义。结论:炎性细胞因子IL-1αI、L-6可能参与了尿石形成过程,α-亚麻酸可能通过抑制炎性细胞炎子的产生而对尿石形成起保护作用。
Objective: To investigate the role of inflammatory cytokines IL-1α, L-1β, IL-6 and TNF-α in the formation of urinary calculus and the effect of α-linolenic acid (α-LNA) on them. Methods: Forty-five male Wistar adult rats were randomly divided into three groups, receiving normal feed plus drinking water (group A), common feed plus chlorite drinking water (group B) and ordinary feed plus inducer drinking water Oil sub-stomach (2 g. Only -1.d-1) (C group). After the eighth week of feeding, the levels of blood cytokines in each group were detected. The control group (group A), IL-6 group (group b), EPA10μmol plus IL-6 group (group C) and EPA50μmol plus IL-6 treatment group were established by using rat mesangial cells (RMC) Group) and ethanol (group e). The content of PGE2 in culture supernatant was detected by radioimmunoassay. Results: The levels of IL-1α and L-6 in group C and group A were significantly lower than those in group B, and there was no significant difference in the levels of IL-1β and TNF-α between the three groups. The content of PGE2 in the cell culture supernatant was significantly lower in groups a, c and d than in group b, while in group d was significantly lower than that in group c. There was no significant difference between groups a and c. CONCLUSION: The inflammatory cytokines IL-1α, L-6 may be involved in the formation of uroliths. Α-linolenic acid may play a protective role in the formation of uroliths by inhibiting the production of inflammatory cytokines.
