目的本研究旨在检测p27Kip1蛋白在鼻咽癌组织中的表达,探讨它在鼻咽癌发生发展中的作用机制,为阐明鼻咽癌发生的分子机制提供实验依据,为鼻咽癌的诊断和预后的评估提供新指标。方法收集广州医学院第二附属医院病理科2002年~2003年存档的60例手术活检切取的鼻咽癌组织组织蜡块,与30例非肿瘤鼻咽组织石蜡标本作比较。应用免疫组织化学技术(SABC)检测鼻咽癌组织中p27Kip1蛋白的表达。结果检测发现p27Kip1蛋白在鼻咽癌细胞核和细胞浆中均有表达。在细胞核表达阳性率为46.7%,非肿瘤鼻咽组织胞核表达为86.7%,前者明显降低,p<0.01;而在细胞浆中表达丰富,阳性率为68.3%,后者为20%,显著高于非肿瘤鼻咽组织的胞浆表达,p<0.01。鼻咽癌中p27Kip1蛋白存在不同于非肿瘤鼻咽组织的胞核和胞浆表达与定位,可见p27Kip1蛋白胞核表达与临床病理的关系更为密切。结论与非肿瘤鼻咽组织相比,p27Kip1蛋白为细胞核低表达、细胞浆高表达。鼻咽癌中p27Kip1蛋白存在不同于非肿瘤鼻咽组织的错误的核浆定位,其在细胞核中表达的减少或丢失可能与鼻咽癌的恶性发展有关。研究提示p27Kip1蛋白的异常表达和定位可能涉及鼻咽癌的分期和转移等;p27Kip1蛋白核表达可作为预测鼻咽癌病期和转移的新指标。
Objective To investigate the expression of p27Kip1 protein in nasopharyngeal carcinoma and to explore its mechanism in the occurrence and development of nasopharyngeal carcinoma and to provide experimental basis for elucidating the molecular mechanism of nasopharyngeal carcinoma, Prognosis assessment provides new indicators. Methods Sixty paraffin-embedded sections of nasopharyngeal carcinoma tissue collected from biopsy specimens collected from 2002 to 2003 in the Second Affiliated Hospital of Guangzhou Medical College were collected for comparison with 30 non-tumor nasopharyngeal paraffin specimens. The expression of p27Kip1 in nasopharyngeal carcinoma tissues was detected by immunohistochemistry (SABC). Results The results showed that p27Kip1 protein was expressed in nucleus and cytoplasm of NPC cells. The positive rate was 46.7% in the nucleus and 86.7% in the non-tumor nasopharyngeal tissue, the former was significantly decreased (p <0.01), while the latter was abundant in the cytoplasm with a positive rate of 68.3% and the latter 20% Higher than non-tumor nasopharyngeal tissue cytoplasmic expression, p <0.01. The expression of p27Kip1 protein in nasopharyngeal carcinoma was different from that in non-tumorous nasopharyngeal tissues. The expression of p27Kip1 was more closely related to the clinical pathology. Conclusion Compared with non-tumor nasopharyngeal tissue, p27Kip1 protein is low in nucleus and high in cytoplasm. The aberrant nuclear plasma localization of p27Kip1 in nasopharyngeal carcinoma is different from that of non-tumor nasopharyngeal tissues. The decrease or loss of its expression in the nucleus may be related to the malignant development of nasopharyngeal carcinoma. The study suggests that abnormal expression and localization of p27Kip1 may involve in the staging and metastasis of nasopharyngeal carcinoma. The expression of p27Kip1 may serve as a new index for predicting the stage and metastasis of nasopharyngeal carcinoma.
